How do I access eBooks or Presentation Plus from Cambridge One Desktop App?

To log in to the app you need to have a Cambridge One account. If you don’t have one, please go to www.cambridgeone.org and create one. You can find instructions in the ‘How do I create an account?’ section of these FAQs if you need them.
Once you have downloaded and launched the Cambridge One app you’ll be redirected to the login page. Use your regular login details to access your account with the app.
If you have already activated an eBook or a Cambridge One Presentation Plus product you will be able to access them now. We recommend that you download them to the app so that you can use them when you’re offline too.
Now you can start working offline. When finished with your work you can synchronise your changes (notes, drawings or last visited page) by connecting to the internet.
You can close the app by clicking ‘Exit’ in the top corner of the app’s frame (“x”). (It will close automatically within one hour if you forget.)
Note that closing the app is not the same as logging out. Closing the app will keep you as an identified user, and you’ll be able to access your downloaded content again if offline. If you log out, the next time you want to access your downloaded content and work on it offline, you will need first to go online and log in again. Once logged in, you can continue working offline.

Presentation Plus is classroom presentation software that can be used with an interactive whiteboard or with a computer and a projector . It allows teachers to display the pages and conduct interactive activities from the Student’s Book. It is also possible to present the Workbook, play Class Audio and the Video Program, display scripts and answer keys, play classroom games, and print out the Teacher’s Edition pages, Video Resource Book activities and teaching notes. In addition, the tools in Presentation Plus allow teachers to zoom in, make notes on pages, and add custom links to other material.

30 Tips: How to Make Good PowerPoint Presentation Designs Fast in 2024
A few tried and true tips can help you speed up your PowerPoint presentation design. Check out 30 of my favorite PowerPoint tips to do just that. Each of these give you PowerPoint slideshow help to create good PowerPoint slides:
1. How Do You Give a Memorable PPT Presentation?
If you're learning the top PowerPoint presentation tips and tricks, you're probably asking yourself: how do I give a presentation that won't be forgotten?
We all want to be remembered. The best PowerPoint slideshow help to make a mark on the audience. There are tried-and-true ways to do just that, and expert Neil Tomlinson shares expertise on being remembered:
Get your main point into the presentation as early as possible (this avoids any risk of audience fatigue or attention span waning), then substantiate your point with facts, figures etc and then reiterate your point at the end in a ‘Summary’.
2. Practice Makes Perfect
Also, don’t forget to practice your presentation. Go through your slide deck a few times to make sure you know it like the back of your hand when the big day arrives. Doing so helps you feel more confident. It'll reduce any anxiety and nervousness you might feel as the presentation day approaches.

Once slides are ready, practice one slide at a time aloud until you feel like you know it and like the flow of speech. Be willing to change anything that does not feel in flow. At the end of learning all your slides, practice the whole talk.

3. Adapt Your Presentation to the Audience
Let's say that you're a seasoned presenter with a pretty standard set of presentation topics. Maybe you're an expert in your field, and you're asked to give a PPT presentation frequently on similar topics.
That's the value of being an expert. You might have a standard spiel that you give your audiences, and your content won't totally change from one presentation to another. That's why it helps to make only slight tweaks to adapt your presentation to each audience.
Leading presentation expert Suzannah Baum offered up this advice:
Different audiences will have different needs and different challenges, which requires me to re-sequence the slides, or create new ones. I tend to do a lot of research on my audiences – via surveys, interviews, and conversations with the hiring manager – to help me better understand what information would be most relevant to them.
How do you adapt to your audience? Here are a few more tips:
- Learn about them. If you're asked to speak, talk to the curator of the presentation to learn more about the audience and their background.
- Ask about them! With contact details, send out a survey or a response link to ask for feedback and preparation info. Ask leading questions like "what do you want to learn?"
- Consider the environment. If you're presenting via Zoom, your style will differ from presenting in person. The key is to acknowledge the difference and adapt to your environment.
Learn everything you can about your audience. Learning how to make a presentable PowerPoint is all about thinking of the recipient, not the presenter!
4. Use a Custom Font
A PowerPoint presentation tip that'll make your slideshow more interesting and more engaging is to use a custom font.
Fonts set the tone for your presentation. So, when you use a premium font, you’re opting for a high-quality font while also adding a personal or creative touch.
When choosing a font, remember that you want everyone to read your text easily.
5. Use Contrast
One PowerPoint trick is to use contrast to make some of your text stand out or make it easier to read.
If you’re putting text over an image on our PowerPoint slide, you may need to use a white box with black text in it to make your text easier to read. You can also use contrasting colors to highlight important text.
6. Avoid Too Many Animations
Another PowerPoint tip is to avoid having too many animations or transitions.
When you've got too many animations, it can be distracting to the audience. It’s not only distracting, but it's unprofessional.
It’s best to stick to one or two animations throughout your presentation. Also, if you've got any animations in your presentation, make sure to test them to see if they work before presenting.
7. Add Audio
Include audio on a slide on PowerPoint to increase audience engagement. Audio can be anything from fun sound effects to interview clips. You can even add an audio clip of your voice.

8. Use the Rule of Three
One PowerPoint tip and trick is to follow the rules of PowerPoint.
One of those rules is the rule of three. It's where you start by dividing your presentation into thirds. Everything should come in thirds, so if you use bullet points, you should only have three. If you use icons, you should only have three.

9. Use a Custom PPT Theme Design
Above all, consistently use custom PowerPoint themes. Microsoft has built-in themes that you can use for free, sure. But the premium themes that are on Envato Elements are a major step-up from PowerPoint's built-in themes.

10. Make Use of Charts and Graphs
Illustrate your data with the use of charts and graphs. Not only will you be able to make your presentation more visually appealing, but you'll also help your audience remember the information better.
Many PowerPoint templates already include chart and graph elements. Easily customize them to make your data and stats more interesting and easier to understand.

When it comes to visualizing data in presentations, we have to remember that our audience does not need all the fine details of the data - they need the main takeaway and we need to make sure that's evident to them when looking at the slide.
11. Use the Built-in Slide Layouts
Inside of PowerPoint themes, you'll find layouts , which are custom slide designs.
Most themes include a selection of content layouts that you can use as a starting point for your own slide designs. You can leverage slide master PowerPoint 2024 designs with the help of layouts.
Layouts are like a starting point for your PowerPoint presentation slides. They contain combinations of placeholders for text boxes, images, and more.
Instead of clicking and drawing individual objects onto the slide, use one of these layouts to start your slide off. It's one of the top PowerPoint presentation tips and tricks to save time.
12. Align Text Consistently
When you're working with text on your slide, it helps to ensure that it aligns consistently. Keeping your text aligned in the same orientation really makes a slide look clean.
In the example below, I've basically got three text boxes:
- list of bulleted points
Notice that all this text is aligned left.
Aligning text was the " aha " moment that I learned when I started studying slide design. It's one of those steps that makes a slide look much neater and professional, so keep it in mind when designing.
13. Make Your Exports User-Friendly
No matter how great your PowerPoint presentation slides look, you need to think about how your user will use the presentation file.
Any of these are likely scenarios if you're regularly sending presentations to other users:
- The viewer may not have PowerPoint installed on their computer.
- The recipient may be using a version of PowerPoint that renders the presentation differently.
- Maybe you don't want the user to be able to make any edits or see your notes in the presentation file.
In this case, my favorite tip is to export the presentation as a PDF. To do that, go to File > Export > Create PDF , and then save your presentation as a PDF.
This is sure to help most of your users see the presentation just the way you intended.
14. Try a Different Color Scheme
Many PowerPoint themes have more than one color scheme that you can apply to your presentation. On the Design tab, click on the drop-down next to Themes to try out a different color scheme.
Typically, these will restyle your entire presentation. Premium themes that you might get from Envato Elements, for example, may have many versions inside the original presentation zip file.
15. Edit Slide Masters for Consistency
The slide master controls the design for your PowerPoint slide. Instead of making the same change to each slide, apply a change to a slide master. It'll affect all the PowerPoint presentation slides that use the same master.
It's ideal to apply a logo to the slide master itself, for example. This keeps the logo the same size and in the same position on each slide.
To do that, go to View > Slide Master. On the right side, you're likely to see a variety of slide masters that control designs for many slides. Drop the elements that you want to remain consistent onto one of the slide masters.
16. Use the Alignment Feature
PowerPoint presentation slides look better when the objects on them are in line with one another. There's a certain visual rhythm that occurs when objects line up in the center or along certain boundary lines.
When you start dragging objects on your slide, you'll see guiding lines that pop up. These are very intuitive, and you'll likely notice that they help you line up your objects. You might seem them pop up when you've got a box that's equidistant between two other objects on the slide, for example.
This is one of the best tricks for improving the look of your PowerPoint slide. Spend some time making sure that your key elements line up cohesively.
17. Use Stock Assets
Earlier, I mentioned using Envato Elements to grab PowerPoint themes. But there's more that comes with an Envato Elements subscription for presentations.

18. Reduce Your Content
There's nothing that makes an audience tune out faster than being overloaded with slide content. Sometimes we try to make so many points that the audience misses all of them due to information overload.
Less is truly more. When you cut the weaker points of your presentation, the audience's attention will follow your key points accordingly.
It seems like cheating, but one of the best steps that you can take for your slide is to simply reduce the number of items that are on it. Convert some of your typed points to things you'll speak verbally.
19. Rethink Your Slide Order
Sometimes, I find that my presentations are out of order. I might spend too much time explaining my decision before I get to the conclusion.
In these cases, I like to use Slide Sorter View to re-sequence the slides in my presentation. To access this view, go to View > Slide Sorter on PowerPoint's ribbon.
From Slide Sorter view, you've got a top-down view of all the slides in your presentation deck. It sometimes becomes obvious that the slides can be reordered into a better sequence from this view.
20. Use PowerPoint Animations
One of my favorite PowerPoint presentation tips is to complement your major points with a bit of animation. Using animation can bring a key point onto your slide with style!

21. Invite Collaborators
Building a presentation often benefits from a second set of eyes. That's why it helps so much to invite a collaborator to work with you side-by-side in Microsoft PowerPoint.
Pushing your presentation up to OneDrive and inviting collaborators is easy. Thanks to the cloud-based approach, more than one user can edit a slide deck in real time. Learn how to do that in the tutorial below:
22. Add Supporting Video Clips
Building impactful presentations is all about adding other perspectives and angles to the content. One of my favorite ways to do that is to add a video clip. Maybe that's a production that you built on your own or found on sites like YouTube.

23. Use Infographic Templates
More presentations than ever will feature visuals that tell stories with data. But it's easy for an audience become overwhelmed with data.
That's where infographics come into play. Learn to use them in PowerPoint in the tutorial below:
24. Use Impactful Closing Techniques
I've sat through many presentations in my life. I can only remember a few that really stick out, thanks to techniques that highlighted key points. You need PowerPoint tips and tricks that help leave your audience with an impact.
To do just that, make sure you use some of the techniques highlighted in the article below:

25. Include Data in the Appendix
Many PowerPoint presentations include data in the form of charts and graphs. That means that you'll condense specifics into a few easy-to-follow charts.
But what if your audience wants more of the backing details? Maybe they want to validate and review the detail for themselves. In that case, a set of appendix slides with extra data is sure to help.
Appendix slides are included at the end of a presentation deck for backup purposes. You might not present them, but your audience is certain to appreciate that you included them. That helps your presentation continue to be useful even after you leave the room.
Here's a great tip from: pro presenter Graeme Thomas of Johnny F Designs:
If (my clients) are sending the deck straight to clients however, I would then put all the information on the slides but will often use more slides so that they aren't too cluttered. In cases where there is a lot of content, like financial statements, I would use appendix slides.
Including an appendix helps your audience understand data without overwhelming them with that data. Follow these tips so that you get the best of both worlds.
26. Alternate Between Solid Color and White Slides
Alternating between solid color and slides with a white background can produce an interesting visual effect and engage your audience. You can use the solid-colored slides to signify a new section in your presentation.
Not to mention, solid-colored slides are the perfect way to re-enforce your brand colors and build your brand recognition.
27. Present Information With Maps

How to prepare presentation slides
Tips, tricks, and a shortcut to getting your next slide deck ready

A good slide deck is the foundation of an informative and engaging presentation. That’s because without well-designed slides, the audience has no visual way to connect the dots in a speech .
Unfortunately, many presenters treat their slides as speaking notes. Worse, they stuff them with irrelevant visuals and make painfully inconsistent formatting choices. All of these gaffes detract from the substance of the presentation and shorten viewers’ already fleeting attention spans.
Creating a slide deck that supports information delivery during the speech demands two actions on your part. First, you need to draft slide content and map it to the deck , so that each slide delivers a key chunk of information. Second, you must use proven design principles to make the slides legible, captivating, and memorable . Below, we’ll walk you through every part of this process — and show you an effective way to prepare presentation-ready slides quickly with AI.
Get your content ready and map it out to the slide deck
Before you start working with a presentation maker like Google Slides or PowerPoint, it’s a good idea to prepare your content and distribute it strategically between the slides .

- Map the content to the slide deck
Now that you have a content outline, consider how it will line up with the slide deck. The 3-2-1 method offers this helpful guidance for planning slides based on a content outline:
- 3 takeaways: Each slide should leave the spectators with 3 important takeaways.
- 2 minutes: You should not spend more than 2 minutes on a single slide.
- 1 idea: Each slide should communicate a single concept or idea.
Using the 3-2-1 method, you can transpose the content outline onto the slides. At this point, you’ll know how many slides are in the presentation . Following the “2 minutes” guideline, you can also calculate how much time the entire presentation will take. This is a good time to gauge whether 2 minutes is enough — or too much — for the ideas and takeaways you will cover in each slide, and adjust the content accordingly.

Even an inattentive spectator should be able to grasp the slide’s main concept at a glance — even if they’re not paying attention to your speech or reading every single bullet point.
Two vital steps will ensure that the slide layouts, backgrounds, and graphics communicate your ideas instead of detracting from them:
- Avoid information overload: Packing the slides with sizable chunks of data and long text passages will cost you your viewers’ attention.

Don’t overload the slides with information
We’ve gone over the 3:2:1 rule above, but there’s a bit more nuance to actually adding content to slides. You may be tempted to write out as much detail as possible to explain each of the ideas and key takeaways. But know this: Nobody in your audience will read long walls of text .
So instead, try to distill all the necessary information into bite-sized pieces . An easy way to do this is to state each idea in the slide header, then present the takeaways as succinct bullet points or text columns:
As for the supporting details? Well, your speech should expound these — they don’t belong on the slides themselves (although you can tuck them into the speaking notes — more on this below).
Slide formatting should be clear and consistent
Even concise, compelling slide content can be lost on viewers if it’s formatted poorly and inconsistently throughout the deck . To avoid designing chaotic slides that distract viewers from the presentation, use consistent fonts, sizes, and colors for different written elements. The formatting should depict the hierarchy of information clearly — your viewers must be able to discern a slide’s header, subheaders, bullet lists, and captions at a single glance:
At the same time, pay attention to the padding between the various elements as well as the slide’s margins. The spacing, including margins, should be even, centered, and uniform from one slide to the next.
The slide deck’s visual design will help you tell your story
Slide background and typography colors communicate a sentiment to the viewers, and contribute to viewer engagement — especially when the subject matter gets a bit dry.
Start by assembling the slideshow’s color scheme. Pick several colors that align with the frame of mind your presentation should evoke — you’ll use them consistently — but in different combinations if you like — in the slide deck.
Make sure that the colors promote legibility through strong contrast — the text should stand out clearly on the slide’s background. Add relevant graphics to the slides
Even with masterfully coordinated colors, the slides will fall flat if they rely on text alone to tell the story. That’s why you should sprinkle relevant graphics throughout the presentation (sparingly). Images, videos, and other graphic assets not only aid information delivery, they also inject a bit of fun into the slideshow.
Likewise, GIFs and memes provide much needed comic relief — especially in info-rich slide decks that are approaching the cognitive overload territory (a death sentence for viewers’ attention spans).
That said, the graphics you add to a slide should be highly relevant to the content . Otherwise, they’ll detract from your message and fail at their task of invigorating the presentation. If using comedic elements like GIFs, insert them strategically into parts of the deck where they’ll capture the sentiment built up by your speech.

Prepare presentation notes for yourself
As we’ve said above, reading the slide content during the speech is a good way to bore your spectators to death. But while this is a big no-no, nothing precludes you from jotting down your talking points for each slide — these will appear only on your computer screen only, and the audience will not see them.

As with slide content, you don’t want to write out walls of text in your presentation notes. If you do, your eyes will be glued to the computer for the duration of the speech. Instead, write out succinct bullet points that expound each takeaway, and these will help guide your speech while letting you interact with the viewers.

Ask for a peer review of the slide deck before you present
You may think that you’ve put together a compelling slide deck, but your proximity to the slideshow may obscure certain nuances or ways to make the slides more effective. Since two heads are usually better than one, consider getting a colleague or friend to review the slides. Ask them to glance at each slide quickly and tell you what the main idea and key takeaways are — if they’re absorbing the information well, then you’ve done a great job preparing the deck.

New longer-term data from the MARIPOSA study confirm superior outcomes of chemotherapy-free RYBREVANT ® plus LAZCLUZE™ regimen compared to osimertinib monotherapy as first-line therapy
Results from an interim analysis featured in late-breaker oral presentation at WCLC
SAN DIEGO , Sept. 8, 2024 /PRNewswire/ -- Johnson & Johnson (NYSE: JNJ) today announced longer follow-up data from the landmark Phase 3 MARIPOSA study which showed first-line treatment with RYBREVANT ® (amivantamab-vmjw) combined with LAZCLUZE™ (lazertinib) provided consistent benefit across long-term outcomes compared to osimertinib monotherapy in adult patients with advanced non-small cell lung cancer (NSCLC) with epidermal growth factor receptor ( EGFR ) exon 19 deletions (ex19del) or L858R substitution mutations. The data show a strong and improving overall survival (OS) trend favoring RYBREVANT ® plus LAZCLUZE™ at approximately three years of follow-up. These results were presented in a late-breaking oral presentation at the International Association for the Study of Lung Cancer (IASLC) 2024 World Conference on Lung Cancer (WCLC) ( Abstract #1146 ). 1
At three years (a median follow-up of 31.1 months), 61 percent of patients receiving RYBREVANT ® plus LACLUZE™ were alive compared to 53 percent of those treated with osimertinib based on an analysis performed at the request of a health authority (Median OS not estimable vs 37.3 months; hazard ratio [HR], 0.77; [95 percent confidence interval [CI], 0.61-0.96]; nominal P =0.019). Overall survival will continue to be assessed with longer term follow-up as a key secondary endpoint. The primary efficacy outcome measure was progression-free survival (PFS) as assessed by blinded independent central review (BICR). 1
"By combining the multi-targeted mechanism of RYBREVANT with LAZCLUZE, a central nervous system-penetrant third-generation tyrosine kinase inhibitor, we are advancing a chemotherapy-free regimen for the first-line treatment of patients with EGFR -mutant NSCLC. This approach blocks EGFR and MET pathways and leverages the immune system, offering patients an opportunity for prolonged benefits," said Shirish M. Gadgeel, M.D., Chief of Division of Hematology and Oncology, Associate Director at Henry Ford Cancer Institute and presenting author.* "Even more encouraging is the marked improvement in the hazard ratio and the ongoing separation of survival curves, showing an eight percent improvement at three years for RYBREVANT plus LAZCLUZE compared to osimertinib. This supports the long-term benefit of the combination as a first-line treatment option in this setting."
Results further showed RYBREVANT ® plus LAZCLUZE™ demonstrated a trend toward improved central nervous system disease control compared to osimertinib at three years (HR, 0.82; [95 percent CI, 0.62-1.09]; nominal P =0.165). At the three-year landmark, intracranial PFS was double for RYBREVANT ® plus LAZCLUZE™ versus osimertinib (38 percent vs 18 percent, respectively). More patients remained on treatment at three years with the RYBREVANT ® combination compared to osimertinib (40 percent vs 29 percent, respectively; HR, 0.80; [95 percent CI, 0.68-0.96]; nominal P =0.014). Additionally, more patients receiving RYBREVANT ® and LAZCLUZE™ at the three-year follow-up had not started a subsequent therapy versus osimertinib (45 percent vs 32 percent, respectively; HR, 0.77; [95 percent CI, 0.65-0.93]; nominal P =0.005). Progression-free survival after first subsequent therapy was 57 percent for the RYBREVANT ® combination compared to 49 percent for osimertinib (HR, 0.73; [95 percent CI, 0.59-0.91]; nominal P =0.004). 1

As previously reported in the MARIPOSA study, the safety profile was consistent with the safety profiles of the individual treatments. The rate of discontinuation of all study treatments due to treatment-related adverse events for RYBREVANT ® plus LAZCLUZE™ was 10 percent. The rate of interstitial lung disease (including pneumonitis) was less than three percent in both arms. 2
In August 2024, RYBREVANT ® combined with LAZCLUZE™ was approved following a Priority Review by the U.S. Food and Drug Administration as a first-line therapy for patients with EGFR -mutated NSCLC based on the favorable efficacy and safety profile demonstrated in this study.
About the MARIPOSA Study
MARIPOSA ( NCT04487080 ), which enrolled 1,074 patients, is a randomized, Phase 3 study evaluating RYBREVANT ® in combination with LAZCLUZE™ versus osimertinib and versus LAZCLUZE™ alone in first-line treatment of patients with locally advanced or metastatic NSCLC with EGFR ex19del or L858R substitution mutations. The primary endpoint of the study is PFS (using RECIST v1.1 guidelines) as assessed by BICR. Secondary endpoints include OS, overall response rate (ORR), duration of response (DOR), second progression-free survival (PFS2) and intracranial PFS. 3
About RYBREVANT ®
RYBREVANT ® (amivantamab-vmjw), a fully-human bispecific antibody targeting EGFR and MET with immune cell-directing activity, is approved in the U.S. , Europe , and in other markets around the world as monotherapy for the treatment of adult patients with locally advanced or metastatic NSCLC with EGFR exon 20 insertion mutations, as detected by an FDA-approved test, whose disease has progressed on or after platinum-based chemotherapy. 4
RYBREVANT ® is approved in the U.S. , Europe and in markets around the world in combination with chemotherapy (carboplatin and pemetrexed) for the first-line treatment of adult patients with locally advanced or metastatic NSCLC with EGFR exon 20 insertion mutations, as detected by an FDA-approved test.
RYBREVANT ® is approved in the U.S. in combination with LAZCLUZE™ (lazertinib) for the first-line treatment of adult patients with locally advanced or metastatic NSCLC with EGFR exon 19 deletions or L858R substitution mutations, as detected by an FDA-approved test. A marketing authorization application (MAA) and type II extension of indication application were submitted to the European Medicines Agency (EMA) seeking approval of LAZCLUZE™ in combination with RYBREVANT ® based on the MARIPOSA study.
In November 2023, Johnson & Johnson submitted a supplemental Biologics License Application (sBLA) to the U.S. FDA for RYBREVANT ® in combination with chemotherapy for the treatment of patients with EGFR-mutated NSCLC who progressed on or after osimertinib based on the MARIPOSA-2 study. This indication was approved in Europe in August 2024.
In June 2024, Johnson & Johnson submitted a BLA to the U.S. FDA for the subcutaneous formulation of RYBREVANT ® in combination with LAZCLUZE™ for all currently approved or submitted indications of intravenous (IV) RYBREVANT ® in certain patients with NSCLC. A submission for the extension of the RYBREVANT ® marketing authorization (line extension) was also submitted to the EMA seeking approval for this indication.
The NCCN Clinical Practice Guidelines in Oncology (NCCN Guidelines ® ) for NSCLC § prefer next-generation sequencing–based strategies over polymerase chain reaction–based approaches for the detection of EGFR exon 20 insertion variants. The NCCN Guidelines include:
- Amivantamab-vmjw (RYBREVANT ® ) plus chemotherapy as a preferred (Category 1 preferred recommendation) subsequent therapy for patients with locally advanced or metastatic NCSLC with EGFR exon 19 deletions or exon 21 L858R mutations who experienced disease progression after treatment with Osimertinib. 5 †‡
- Amivantamab-vmjw (RYBREVANT ® ) plus carboplatin and pemetrexed as a preferred (Category 1 preferred recommendation) first-line therapy in treatment-naive patients with newly diagnosed advanced or metastatic EGFR exon 20 insertion mutation-positive advanced NSCLC, or as a subsequent therapy option (Category 2A recommendation) for patients that have progressed on or after platinum-based chemotherapy with or without immunotherapy and have EGFR exon 20 insertion mutation-positive advanced NSCLC. 5 †‡

About LAZCLUZE ™
In 2018, Janssen Biotech, Inc., entered into a license and collaboration agreement with Yuhan Corporation for the development of LAZCLUZE™ (marketed as LACLAZA in Korea). LAZCLUZE™ is an oral, third-generation, brain-penetrant EGFR TKI that targets both the T790M mutation and activating EGFR mutations while sparing wild-type EGFR. About Non-Small Cell Lung Cancer
Worldwide, lung cancer is one of the most common cancers, with NSCLC making up 80 to 85 percent of all lung cancer cases. 19,20 The main subtypes of NSCLC are adenocarcinoma, squamous cell carcinoma, and large cell carcinoma. 21 Among the most common driver mutations in NSCLC are alterations in EGFR , which is a receptor tyrosine kinase controlling cell growth and division. 22 EGFR mutations are present in 10 to 15 percent of Western patients with NSCLC with adenocarcinoma histology and occur in 40 to 50 percent of Asian patients. 21,22,23,24,25,26 EGFR ex19del or EGFR L858R mutations are the most common EGFR mutations. 27 The five- year survival rate for all people with advanced NSCLC and EGFR mutations treated with EGFR tyrosine kinase inhibitors (TKIs) is less than 20 percent. 28,29 EGFR exon 20 insertion mutations are the third most prevalent activating EGFR mutation. 30 Patients with EGFR exon 20 insertion mutations have a real-world five-year overall survival (OS) of eight percent in the frontline setting, which is worse than patients with EGFR ex19del or L858R mutations, who have a real-world five-year OS of 19 percent. 31

WARNINGS AND PRECAUTIONS
Infusion-Related Reactions
RYBREVANT ® can cause infusion-related reactions (IRR); signs and symptoms of IRR include dyspnea, flushing, fever, chills, nausea, chest discomfort, hypotension, and vomiting. The median time to IRR onset is approximately 1 hour.
RYBREVANT ® with LAZCLUZE™
RYBREVANT ® in combination with LAZCLUZE™ can cause infusion-related reactions. In MARIPOSA (n=421), IRRs occurred in 63% of patients treated with RYBREVANT ® in combination with LAZCLUZE™, including Grade 3 in 5% and Grade 4 in 1% of patients. The incidence of infusion modifications due to IRR was 54% of patients, and IRRs leading to dose reduction of RYBREVANT ® occurred in 0.7% of patients. Infusion-related reactions leading to permanent discontinuation of RYBREVANT ® occurred in 4.5% of patients receiving RYBREVANT ® in combination with LAZCLUZE™.
RYBREVANT ® with Carboplatin and Pemetrexed
In PAPILLON (n=151), infusion-related reactions occurred in 42% of patients treated with RYBREVANT ® in combination with carboplatin and pemetrexed, including Grade 3 (1.3%) adverse reactions. The incidence of infusion modifications due to IRR was 40%, and 0.7% of patients permanently discontinued RYBREVANT ® .
RYBREVANT ® as a Single Agent
In CHRYSALIS (n=302), IRR occurred in 66% of patients treated with RYBREVANT ® . Among patients receiving treatment on Week 1 Day 1, 65% experienced an IRR, while the incidence of IRR was 3.4% with the Day 2 infusion, 0.4% with the Week 2 infusion, and cumulatively 1.1% with subsequent infusions. Of the reported IRRs, 97% were Grade 1-2, 2.2% were Grade 3, and 0.4% were Grade 4. The median time to onset was 1 hour (range 0.1 to 18 hours) after start of infusion. The incidence of infusion modifications due to IRR was 62% and 1.3% of patients permanently discontinued RYBREVANT ® due to IRR.
Premedicate with antihistamines, antipyretics, and glucocorticoids and infuse RYBREVANT ® as recommended. Administer RYBREVANT ® via a peripheral line on Week 1 and Week 2 to reduce the risk of infusion-related reactions. Monitor patients for signs and symptoms of infusion reactions during RYBREVANT ® infusion in a setting where cardiopulmonary resuscitation medication and equipment are available. Interrupt infusion if IRR is suspected. Reduce the infusion rate or permanently discontinue RYBREVANT ® based on severity.
Interstitial Lung Disease/Pneumonitis
RYBREVANT ® can cause severe and fatal interstitial lung disease (ILD)/pneumonitis.
In MARIPOSA, ILD/pneumonitis occurred in 3.1% of patients treated with RYBREVANT ® in combination with LAZCLUZE™, including Grade 3 in 1.0% and Grade 4 in 0.2% of patients. There was one fatal case (0.2%) of ILD/pneumonitis and 2.9% of patients permanently discontinued RYBREVANT ® and LAZCLUZE™ due to ILD/pneumonitis.
In PAPILLON, Grade 3 ILD/pneumonitis occurred in 2.6% of patients treated with RYBREVANT ® in combination with carboplatin and pemetrexed, all patients required permanent discontinuation.
In CHRYSALIS, ILD/pneumonitis occurred in 3.3% of patients treated with RYBREVANT ® , with 0.7% of patients experiencing Grade 3 ILD/pneumonitis. Three patients (1%) discontinued RYBREVANT ® due to ILD/pneumonitis.
Monitor patients for new or worsening symptoms indicative of ILD/pneumonitis (e.g., dyspnea, cough, fever). For patients receiving RYBREVANT ® in combination with LAZCLUZE™, immediately withhold both drugs in patients with suspected ILD/pneumonitis and permanently discontinue if ILD/pneumonitis is confirmed. For patients receiving RYBREVANT ® as a single agent or in combination with carboplatin and pemetrexed, immediately withhold RYBREVANT ® in patients with suspected ILD/pneumonitis and permanently discontinue if ILD/pneumonitis is confirmed.
Venous Thromboembolic (VTE) Events with Concomitant Use of RYBREVANT ® and LAZCLUZE™
RYBREVANT ® in combination with LAZCLUZE™ can cause serious and fatal venous thromboembolic (VTEs) events, including deep vein thrombosis and pulmonary embolism. The majority of these events occurred during the first four months of therapy.
In MARIPOSA, VTEs occurred in 36% of patients receiving RYBREVANT ® in combination with LAZCLUZE™, including Grade 3 in 10% and Grade 4 in 0.5% of patients. On-study VTEs occurred in 1.2% of patients (n=5) while receiving anticoagulation therapy. There were two fatal cases of VTE (0.5%), 9% of patients had VTE leading to dose interruptions of RYBREVANT ® , and 7% of patients had VTE leading to dose interruptions of LAZCLUZE™; 1% of patients had VTE leading to dose reductions of RYBREVANT ® , and 0.5% of patients had VTE leading to dose reductions of LAZCLUZE™; 3.1% of patients had VTE leading to permanent discontinuation of RYBREVANT ® , and 1.9% of patients had VTE leading to permanent discontinuation of LAZCLUZE™. The median time to onset of VTEs was 84 days (range: 6 to 777).
Administer prophylactic anticoagulation for the first four months of treatment. The use of Vitamin K antagonists is not recommended. Monitor for signs and symptoms of VTE events and treat as medically appropriate.
Withhold RYBREVANT ® and LAZCLUZE™ based on severity. Once anticoagulant treatment has been initiated, resume RYBREVANT ® and LAZCLUZE™ at the same dose level at the discretion of the healthcare provider. In the event of VTE recurrence despite therapeutic anticoagulation, permanently discontinue RYBREVANT ® and continue treatment with LAZCLUZE™ at the same dose level at the discretion of the healthcare provider.
Dermatologic Adverse Reactions
RYBREVANT ® can cause severe rash including toxic epidermal necrolysis (TEN), dermatitis acneiform, pruritus, and dry skin.
In MARIPOSA, rash occurred in 86% of patients treated with RYBREVANT ® in combination with LAZCLUZE™, including Grade 3 in 26% of patients. The median time to onset of rash was 14 days (range: 1 to 556 days). Rash leading to dose interruptions occurred in 37% of patients for RYBREVANT ® and 30% for LAZCLUZE™, rash leading to dose reductions occurred in 23% of patients for RYBREVANT ® and 19% for LAZCLUZE™, and rash leading to permanent discontinuation occurred in 5% of patients for RYBREVANT ® and 1.7% for LAZCLUZE™.
In PAPILLON, rash occurred in 89% of patients treated with RYBREVANT ® in combination with carboplatin and pemetrexed, including Grade 3 (19%) adverse reactions. Rash leading to dose reductions occurred in 19% of patients, and 2% permanently discontinued RYBREVANT ® and 1.3% discontinued pemetrexed.
In CHRYSALIS, rash occurred in 74% of patients treated with RYBREVANT ® as a single agent, including Grade 3 rash in 3.3% of patients. The median time to onset of rash was 14 days (range: 1 to 276 days). Rash leading to dose reduction occurred in 5% of patients, and RYBREVANT ® was permanently discontinued due to rash in 0.7% of patients.
Toxic epidermal necrolysis occurred in one patient (0.3%) treated with RYBREVANT ® as a single agent.
Instruct patients to limit sun exposure during and for 2 months after treatment with RYBREVANT ® or LAZCLUZE™ in combination with RYBREVANT ® . Advise patients to wear protective clothing and use broad-spectrum UVA/UVB sunscreen. Alcohol-free (e.g., isopropanol-free, ethanol-free) emollient cream is recommended for dry skin.
When initiating RYBREVANT ® treatment with or without LAZCLUZE™, administer alcohol-free emollient cream to reduce the risk of dermatologic adverse reactions. Consider prophylactic measures (e.g. use of oral antibiotics) to reduce the risk of dermatologic reactions. If skin reactions develop, start topical corticosteroids and topical and/or oral antibiotics. For Grade 3 reactions, add oral steroids and consider dermatologic consultation. Promptly refer patients presenting with severe rash, atypical appearance or distribution, or lack of improvement within 2 weeks to a dermatologist. For patients receiving RYBREVANT ® in combination with LAZCLUZE™, withhold, dose reduce or permanently discontinue both drugs based on severity. For patients receiving RYBREVANT ® as a single agent or in combination with carboplatin and pemetrexed, withhold, dose reduce or permanently discontinue RYBREVANT ® based on severity.
Ocular Toxicity
RYBREVANT ® can cause ocular toxicity including keratitis, blepharitis, dry eye symptoms, conjunctival redness, blurred vision, visual impairment, ocular itching, eye pruritus, and uveitis.
In MARIPOSA, ocular toxicity occurred in 16% of patients treated with RYBREVANT ® in combination with LAZCLUZE™, including Grade 3 or 4 ocular toxicity in 0.7% of patients. Withhold, reduce the dose, or permanently discontinue RYBREVANT ® and continue LAZCLUZE™ based on severity.
In PAPILLON, ocular toxicity including blepharitis, dry eye, conjunctival redness, blurred vision, and eye pruritus occurred in 9%. All events were Grade 1-2.
In CHRYSALIS, keratitis occurred in 0.7% and uveitis occurred in 0.3% of patients treated with RYBREVANT ® . All events were Grade 1-2.
Promptly refer patients with new or worsening eye symptoms to an ophthalmologist. Withhold, dose reduce or permanently discontinue RYBREVANT ® based on severity.
Embryo-Fetal Toxicity
Based on its mechanism of action and findings from animal models, RYBREVANT ® and LAZCLUZE™ can cause fetal harm when administered to a pregnant woman. Advise females of reproductive potential of the potential risk to the fetus.
Advise female patients of reproductive potential to use effective contraception during treatment and for 3 months after the last dose of RYBREVANT ® .
Advise females of reproductive potential to use effective contraception during treatment with LAZCLUZE™ and for 3 weeks after the last dose. Advise male patients with female partners of reproductive potential to use effective contraception during treatment with LAZCLUZE™ and for 3 weeks after the last dose.
Adverse Reactions
For the 421 patients in the MARIPOSA clinical trial who received RYBREVANT ® in combination with LAZCLUZE™, the most common adverse reactions (≥20%) were rash (86%), nail toxicity (71%), infusion-related reactions (RYBREVANT ® , 63%), musculoskeletal pain (47%), stomatitis (43%), edema (43%), VTE (36%), paresthesia (35%), fatigue (32%), diarrhea (31%), constipation (29%), COVID-19 (26%), hemorrhage (25%), dry skin (25%), decreased appetite (24%), pruritus (24%), nausea (21%), and ocular toxicity (16%). The most common Grade 3 or 4 laboratory abnormalities (≥2%) were decreased albumin (8%), decreased sodium (7%), increased ALT (7%), decreased potassium (5%), decreased hemoglobin (3.8%), increased AST (3.8%), increased GGT (2.6%), and increased magnesium (2.6%).
Serious adverse reactions occurred in 49% of patients who received RYBREVANT ® in combination with LAZCLUZE™. Serious adverse reactions occurring in ≥2% of patients included VTE (11%), pneumonia (4%), ILD/pneumonitis and rash (2.9% each), COVID-19 (2.4%), and pleural effusion and infusion-related reaction (RYBREVANT ® ) (2.1% each). Fatal adverse reactions occurred in 7% of patients who received RYBREVANT ® in combination with LAZCLUZE™ due to death not otherwise specified (1.2%); sepsis and respiratory failure (1% each); pneumonia, myocardial infarction, and sudden death (0.7% each); cerebral infarction, pulmonary embolism (PE), and COVID-19 infection (0.5% each); and ILD/pneumonitis, acute respiratory distress syndrome (ARDS), and cardiopulmonary arrest (0.2% each).
For the 151 patients in the PAPILLON clinical trial who received RYBREVANT ® in combination with carboplatin and pemetrexed, the most common adverse reactions (≥20%) were rash (90%), nail toxicity (62%), stomatitis (43%), infusion-related reaction (42%), fatigue (42%), edema (40%), constipation (40%), decreased appetite (36%), nausea (36%), COVID-19 (24%), diarrhea (21%), and vomiting (21%). The most common Grade 3 to 4 laboratory abnormalities (≥2%) were decreased albumin (7%), increased alanine aminotransferase (4%), increased gamma-glutamyl transferase (4%), decreased sodium (7%), decreased potassium (11%), decreased magnesium (2%), and decreases in white blood cells (17%), hemoglobin (11%), neutrophils (36%), platelets (10%), and lymphocytes (11%).
Serious adverse reactions occurred in 37% of patients who received RYBREVANT ® in combination with carboplatin and pemetrexed. Serious adverse reactions in ≥2% of patients included rash, pneumonia, ILD, pulmonary embolism, vomiting, and COVID-19. Fatal adverse reactions occurred in 7 patients (4.6%) due to pneumonia, cerebrovascular accident, cardio-respiratory arrest, COVID-19, sepsis, and death not otherwise specified.
For the 129 patients in the CHRYSALIS clinical trial who received RYBREVANT ® as a single agent, the most common adverse reactions (≥20%) were rash (84%), IRR (64%), paronychia (50%), musculoskeletal pain (47%), dyspnea (37%), nausea (36%), fatigue (33%), edema (27%), stomatitis (26%), cough (25%), constipation (23%), and vomiting (22%). The most common Grade 3 to 4 laboratory abnormalities (≥2%) were decreased lymphocytes (8%), decreased albumin (8%), decreased phosphate (8%), decreased potassium (6%), increased alkaline phosphatase (4.8%), increased glucose (4%), increased gamma-glutamyl transferase (4%), and decreased sodium (4%).
Serious adverse reactions occurred in 30% of patients who received RYBREVANT ® . Serious adverse reactions in ≥2% of patients included pulmonary embolism, pneumonitis/ILD, dyspnea, musculoskeletal pain, pneumonia, and muscular weakness. Fatal adverse reactions occurred in 2 patients (1.5%) due to pneumonia and 1 patient (0.8%) due to sudden death.
LAZCLUZE™ Drug Interactions
Avoid concomitant use of LAZCLUZE™ with strong and moderate CYP3A4 inducers. Consider an alternate concomitant medication with no potential to induce CYP3A4.
Monitor for adverse reactions associated with a CYP3A4 or BCRP substrate where minimal concentration changes may lead to serious adverse reactions, as recommended in the approved product labeling for the CYP3A4 or BCRP substrate.
