Humanities theses are often structured more like a longer-form essay . Just like in an essay, you build an argument to support a central thesis.

Broad thesis (arguable, but difficult to support with evidence) 

Michael Sandel’s arguments about genetic engineering do not take into consideration all the relevant issues.

This is an arguable claim because it would be possible to argue against it by saying that Michael Sandel’s arguments do take all of the relevant issues into consideration. But the claim is too broad. Because the thesis does not specify which “issues” it is focused on—or why it matters if they are considered—readers won’t know what the rest of the essay will argue, and the writer won’t know what to focus on. If there is a particular issue that Sandel does not address, then a more specific version of the thesis would include that issue—hand an explanation of why it is important.  

Arguable thesis with analytical claim 

While Sandel argues persuasively that our instinct to “remake” (54) ourselves into something ever more perfect is a problem, his belief that we can always draw a line between what is medically necessary and what makes us simply “better than well” (51) is less convincing.

This is an arguable analytical claim. To argue for this claim, the essay writer will need to show how evidence from the article itself points to this interpretation. It’s also a reasonable scope for a thesis because it can be supported with evidence available in the text and is neither too broad nor too narrow.  

Arguable thesis with normative claim 

Given Sandel’s argument against genetic enhancement, we should not allow parents to decide on using Human Growth Hormone for their children.

This thesis tells us what we should do about a particular issue discussed in Sandel’s article, but it does not tell us how we should understand Sandel’s argument.

For the sample assignment above, you’ll want to look at books and articles on World War II in general, and Spain’s neutrality in particular.

As you consider your options, you must decide to focus on one aspect of your topic. This means that you cannot include everything you’ve learned about your topic, nor should you go off in several directions. If you end up covering too many different aspects of a topic, your paper will sprawl and be unconvincing in its argument, and it most likely will not fulfull the assignment requirements.

For the sample assignment above, both Spain’s neutrality and World War II are topics far too broad to explore in a paper. You may instead decide to focus on Franco’s role in the diplomatic relationships between the Allies and the Axis , which narrows down what aspects of Spain’s neutrality and World War II you want to discuss, as well as establishes a specific link between those two aspects.

Before you go too far, however, ask yourself whether your topic is worthy of your efforts. Try to avoid topics that already have too much written about them (i.e., “eating disorders and body image among adolescent women”) or that simply are not important (i.e. “why I like ice cream”). These topics may lead to a thesis that is either dry fact or a weird claim that cannot be supported. A good thesis falls somewhere between the two extremes. To arrive at this point, ask yourself what is new, interesting, contestable, or controversial about your topic.

As you work on your thesis, remember to keep the rest of your paper in mind at all times . Sometimes your thesis needs to evolve as you develop new insights, find new evidence, or take a different approach to your topic.

As you read more about Franco’s decisions, you may conclude that Spain’s neutrality in WWII occurred for an entirely personal reason: his desire to preserve his own (and Spain’s) power. Based on this conclusion, you can then write a trial thesis statement to help you decide what material belongs in your paper.

Sometimes you won’t be able to find a focus or identify your “spin” or specific argument immediately. Like some writers, you might begin with a purpose statement just to get yourself going. A purpose statement is one or more sentences that announce your topic and indicate the structure of the paper but do not state the conclusions you have drawn . Thus, you might begin with something like this:

  • This paper will look at modern language to see if it reflects male dominance or female oppression.
  • I plan to analyze anger and derision in offensive language to see if they represent a challenge of society’s authority.

At some point, you can turn a purpose statement into a thesis statement. As you think and write about your topic, you can restrict, clarify, and refine your argument, crafting your thesis statement to reflect your thinking.

As you work on your thesis, remember to keep the rest of your paper in mind at all times. Sometimes your thesis needs to evolve as you develop new insights, find new evidence, or take a different approach to your topic.

Compose a draft thesis statement

If you are writing a paper that will have an argumentative thesis and are having trouble getting started, the techniques in the table below may help you develop a temporary or “working” thesis statement.

Begin with a purpose statement that you will later turn into a thesis statement.

Assignment: Discuss the history of the Reform Party and explain its influence on the 1990 presidential and Congressional election.

Purpose Statement: This paper briefly sketches the history of the grassroots, conservative, Perot-led Reform Party and analyzes how it influenced the economic and social ideologies of the two mainstream parties.

Question-to-Assertion

If your assignment asks a specific question(s), turn the question(s) into an assertion and give reasons why it is true or reasons for your opinion.

Assignment : What do Aylmer and Rappaccini have to be proud of? Why aren’t they satisfied with these things? How does pride, as demonstrated in “The Birthmark” and “Rappaccini’s Daughter,” lead to unexpected problems?

Beginning thesis statement: Alymer and Rappaccinni are proud of their great knowledge; however, they are also very greedy and are driven to use their knowledge to alter some aspect of nature as a test of their ability. Evil results when they try to “play God.”

Write a sentence that summarizes the main idea of the essay you plan to write.

Main idea: The reason some toys succeed in the market is that they appeal to the consumers’ sense of the ridiculous and their basic desire to laugh at themselves.

Make a list of the ideas that you want to include; consider the ideas and try to group them.

  • nature = peaceful
  • war matériel = violent (competes with 1?)
  • need for time and space to mourn the dead
  • war is inescapable (competes with 3?)

Use a formula to arrive at a working thesis statement (you will revise this later).

  • although most readers of _______ have argued that _______, closer examination shows that _______.
  • _______ uses _______ and _____ to prove that ________.
  • phenomenon x is a result of the combination of __________, __________, and _________.

What to keep in mind as you draft an initial thesis statement

Beginning statements obtained through the methods illustrated above can serve as a framework for planning or drafting your paper, but remember they’re not yet the specific, argumentative thesis you want for the final version of your paper. In fact, in its first stages, a thesis statement usually is ill-formed or rough and serves only as a planning tool.

As you write, you may discover evidence that does not fit your temporary or “working” thesis. Or you may reach deeper insights about your topic as you do more research, and you will find that your thesis statement has to be more complicated to match the evidence that you want to use.

You must be willing to reject or omit some evidence in order to keep your paper cohesive and your reader focused. Or you may have to revise your thesis to match the evidence and insights that you want to discuss. Read your draft carefully, noting the conclusions you have drawn and the major ideas which support or prove those conclusions. These will be the elements of your final thesis statement.

Sometimes you will not be able to identify these elements in your early drafts, but as you consider how your argument is developing and how your evidence supports your main idea, ask yourself, “ What is the main point that I want to prove/discuss? ” and “ How will I convince the reader that this is true? ” When you can answer these questions, then you can begin to refine the thesis statement.

Refine and polish the thesis statement

To get to your final thesis, you’ll need to refine your draft thesis so that it’s specific and arguable.

  • Ask if your draft thesis addresses the assignment
  • Question each part of your draft thesis
  • Clarify vague phrases and assertions
  • Investigate alternatives to your draft thesis

Consult the example below for suggestions on how to refine your draft thesis statement.

Sample Assignment

Choose an activity and define it as a symbol of American culture. Your essay should cause the reader to think critically about the society which produces and enjoys that activity.

  • Ask The phenomenon of drive-in facilities is an interesting symbol of american culture, and these facilities demonstrate significant characteristics of our society.This statement does not fulfill the assignment because it does not require the reader to think critically about society.
Drive-ins are an interesting symbol of American culture because they represent Americans’ significant creativity and business ingenuity.
Among the types of drive-in facilities familiar during the twentieth century, drive-in movie theaters best represent American creativity, not merely because they were the forerunner of later drive-ins and drive-throughs, but because of their impact on our culture: they changed our relationship to the automobile, changed the way people experienced movies, and changed movie-going into a family activity.
While drive-in facilities such as those at fast-food establishments, banks, pharmacies, and dry cleaners symbolize America’s economic ingenuity, they also have affected our personal standards.
While drive-in facilities such as those at fast- food restaurants, banks, pharmacies, and dry cleaners symbolize (1) Americans’ business ingenuity, they also have contributed (2) to an increasing homogenization of our culture, (3) a willingness to depersonalize relationships with others, and (4) a tendency to sacrifice quality for convenience.

This statement is now specific and fulfills all parts of the assignment. This version, like any good thesis, is not self-evident; its points, 1-4, will have to be proven with evidence in the body of the paper. The numbers in this statement indicate the order in which the points will be presented. Depending on the length of the paper, there could be one paragraph for each numbered item or there could be blocks of paragraph for even pages for each one.

Complete the final thesis statement

The bottom line.

As you move through the process of crafting a thesis, you’ll need to remember four things:

  • Context matters! Think about your course materials and lectures. Try to relate your thesis to the ideas your instructor is discussing.
  • As you go through the process described in this section, always keep your assignment in mind . You will be more successful when your thesis (and paper) responds to the assignment than if it argues a semi-related idea.
  • Your thesis statement should be precise, focused, and contestable ; it should predict the sub-theses or blocks of information that you will use to prove your argument.
  • Make sure that you keep the rest of your paper in mind at all times. Change your thesis as your paper evolves, because you do not want your thesis to promise more than your paper actually delivers.

In the beginning, the thesis statement was a tool to help you sharpen your focus, limit material and establish the paper’s purpose. When your paper is finished, however, the thesis statement becomes a tool for your reader. It tells the reader what you have learned about your topic and what evidence led you to your conclusion. It keeps the reader on track–well able to understand and appreciate your argument.

Tips for Writing Your Thesis Statement

1. Determine what kind of paper you are writing:

  • An analytical paper breaks down an issue or an idea into its component parts, evaluates the issue or idea, and presents this breakdown and evaluation to the audience.
  • An expository (explanatory) paper explains something to the audience.
  • An argumentative paper makes a claim about a topic and justifies this claim with specific evidence. The claim could be an opinion, a policy proposal, an evaluation, a cause-and-effect statement, or an interpretation. The goal of the argumentative paper is to convince the audience that the claim is true based on the evidence provided.

If you are writing a text that does not fall under these three categories (e.g., a narrative), a thesis statement somewhere in the first paragraph could still be helpful to your reader.

2. Your thesis statement should be specific—it should cover only what you will discuss in your paper and should be supported with specific evidence.

3. The thesis statement usually appears at the end of the first paragraph of a paper.

4. Your topic may change as you write, so you may need to revise your thesis statement to reflect exactly what you have discussed in the paper.

Developing a Research Thesis

A research thesis has most of the same  thesis characteristics as a thesis for a non-research essay. The difference lies in the fact that you will be gathering information and evidence from appropriate, valid sources to support your perspective on a topic or stand on an issue. Yet although your sources provide information that informs your thesis, the thesis ideas should be your own, particular to your personal way of thinking about and analyzing a topic.

The thesis focuses your ideas and information for the research paper. Remember that word "focus." Student writers often make the mistake of forgetting the focus and making the research thesis far too broad in order to include too much research. Yet depth more than breadth is the hallmark of a sophisticated research paper.

Create a working thesis for the research paper by specifying and ordering your categories of information. For example, the following theses offer the writers' main arguments and focus their research by specifying and ordering the reasons for their stance:

  • Competency-based management is practical, logical, and accessible to managers and workers.
  • Workforce training in basic skills is now necessary because of the lower reading and writing levels of the workforce, the fact that a person now entering the workforce will change jobs many times, the shift in jobs from the manufacturing to the service sectors, and the more comprehensive, abstract nature of those service jobs.

As you can see, a research thesis is your proposed answer to your research question, which you finalize only after completing the research. (It's okay to modify and revise the working thesis as you research more about the topic or issue.)

Developing a good working thesis, just like developing a good research question (researchable: neither too broad nor too narrow) is an important research skill.

This article explains the meaning of research methodology and the purpose and importance of writing a research methodology section or chapter for your thesis paper. It discusses what to include and not include in a research methodology section, the different approaches to research methodology that can be used, and the steps involved in writing a robust research methodology section.

What is a thesis research methodology?

A thesis research methodology explains the type of research performed, justifies the methods that you chose   by linking back to the literature review , and describes the data collection and analysis procedures. It is included in your thesis after the Introduction section . Most importantly, this is the section where the readers of your study evaluate its validity and reliability.

What should the research methodology section in your thesis include?

  • The aim of your thesis
  • An outline of the research methods chosen (qualitative, quantitative, or mixed methods)
  • Background and rationale for the methods chosen, explaining why one method was chosen over another
  • Methods used for data collection and data analysis
  • Materials and equipment used—keep this brief
  • Difficulties encountered during data collection and analysis. It is expected that problems will occur during your research process. Use this as an opportunity to demonstrate your problem-solving abilities by explaining how you overcame all obstacles. This builds your readers’ confidence in your study findings.
  • A brief evaluation of your research explaining whether your results were conclusive and whether your choice of methodology was effective in practice

What should not be included in the research methodology section of your thesis?

  • Irrelevant details, for example, an extensive review of methodologies (this belongs in the literature review section) or information that does not contribute to the readers’ understanding of your chosen methods
  • A description of basic procedures
  • Excessive details about materials and equipment used. If an extremely long and detailed list is necessary, add it as an appendix

Types of methodological approaches

The choice of which methodological approach to use depends on your field of research and your thesis question. Your methodology should establish a clear relationship with your thesis question and must also be supported by your  literature review . Types of methodological approaches include quantitative, qualitative, or mixed methods. 

Quantitative studies generate data in the form of numbers   to count, classify, measure, or identify relationships or patterns. Information may be collected by performing experiments and tests, conducting surveys, or using existing data. The data are analyzed using  statistical tests and presented as charts or graphs. Quantitative data are typically used in the Sciences domain.

For example, analyzing the effect of a change, such as alterations in electricity consumption by municipalities after installing LED streetlights.

The raw data will need to be prepared for statistical analysis by identifying variables and checking for missing data and outliers. Details of the statistical software program used (name of the package, version number, and supplier name and location) must also be mentioned.

Qualitative studies gather non-numerical data using, for example, observations, focus groups, and in-depth interviews.   Open-ended questions are often posed. This yields rich, detailed, and descriptive results. Qualitative studies are usually   subjective and are helpful for investigating social and cultural phenomena, which are difficult to quantify. Qualitative studies are typically used in the Humanities and Social Sciences (HSS) domain.

For example, determining customer perceptions on the extension of a range of baking utensils to include silicone muffin trays.

The raw data will need to be prepared for analysis by coding and categorizing ideas and themes to interpret the meaning behind the responses given.

Mixed methods use a combination of quantitative and qualitative approaches to present multiple findings about a single phenomenon. T his enables triangulation: verification of the data from two or more sources.

Data collection

Explain the rationale behind the sampling procedure you have chosen. This could involve probability sampling (a random sample from the study population) or non-probability sampling (does not use a random sample).

For quantitative studies, describe the sampling procedure and whether statistical tests were used to determine the  sample size .

Following our example of analyzing the changes in electricity consumption by municipalities after installing LED streetlights, you will need to determine which municipal areas will be sampled and how the information will be gathered (e.g., a physical survey of the streetlights or reviewing purchase orders).

For qualitative research, describe how the participants were chosen and how the data is going to be collected.

Following our example about determining customer perceptions on the extension of a range of baking utensils to include silicone muffin trays, you will need to decide the criteria for inclusion as a study participant (e.g., women aged 20–70 years, bakeries, and bakery supply shops) and how the information will be collected (e.g., interviews, focus groups, online or in-person questionnaires, or video recordings) .

Data analysis

For quantitative research, describe what tests you plan to perform and why you have chosen them. Popular data analysis methods in quantitative research include:

  • Descriptive statistics (e.g., means, medians, modes)
  • Inferential statistics (e.g., correlation, regression, structural equation modeling)

For qualitative research, describe how the data is going to be analyzed and justify your choice. Popular data analysis methods in qualitative research include:

  • Qualitative content analysis
  • Thematic analysis
  • Discourse analysis
  • Narrative analysis
  • Grounded theory
  • Interpretative phenomenological analysis (IPA)

Evaluate and justify your methodological choices

You need to convince the reader that you have made the correct methodological choices. Once again, this ties back to your thesis question and  literature review . Write using a persuasive tone, and use  rhetoric to convince the reader of the quality, reliability, and validity of your research.

Ethical considerations

  • The young researcher should maintain objectivity at all times
  • All participants have the right to privacy and anonymity
  • Research participation must be voluntary
  • All subjects have the right to withdraw from the research at any time
  • Consent must be obtained from all participants before starting the research
  • Confidentiality of data provided by individuals must be maintained
  • Consider how the interpretation and reporting of the data will affect the participants

Tips for writing a robust thesis research methodology

  • Determine what kind of knowledge you are trying to uncover. For example, subjective or objective, experimental or interpretive.
  • A thorough literature review is the best starting point for choosing your methods.
  • Ensure that there is continuity throughout the research process. The authenticity of your research depends upon the validity of the research data, the reliability of your data measurements, and the time taken to conduct the analysis.
  • Choose a research method that is achievable. Consider the time and funds available, feasibility, ethics, and access and availability of equipment to measure the phenomenon or answer your thesis question correctly.
  • If you are struggling with a concept, ask for help from your supervisor, academic staff members, or fellow students.

A thesis methodology justifies why you have chosen a specific approach to address your thesis question. It explains how you will collect the data and analyze it. Above all, it allows the readers of your study to evaluate its validity and reliability.

Review Checklist

Introduce your methodological approach , for example, quantitative, qualitative, or mixed methods.

Explain why your chosen approach is relevant to the overall research design and how it links with your  thesis question.

Justify your chosen method and why it is more appropriate than others.

Provide background information on methods that may be unfamiliar to readers of your thesis.

Introduce the tools that you will use for data collection , and explain how you plan to use them (e.g., surveys, interviews, experiments, or existing data).

Explain how you will analyze your results. The type of analysis used depends on the methods you chose. For example, exploring theoretical perspectives to support your explanation of observed behaviors in a qualitative study or using statistical analyses in a quantitative study.

Mention any research limitations. All studies are expected to have limitations, such as the sample size, data collection method, or equipment. Discussing the limitations justifies your choice of methodology despite the risks. It also explains under which conditions the results should be interpreted and shows that you have taken a holistic approach to your study.

What is the difference between methodology and methods? +

Methodology  refers to the overall rationale and strategy of your thesis project. It involves studying the theories or principles behind the methods used in your field so that you can explain why you chose a particular method for your research approach.  Methods , on the other hand, refer to how the data were collected and analyzed (e.g., experiments, surveys, observations, interviews, and statistical tests).

What is the difference between reliability and validity? +

Reliability refers to whether a measurement is consistent (i.e., the results can be reproduced under the same conditions).  Validity refers to whether a measurement is accurate (i.e., the results represent what was supposed to be measured). For example, when investigating linguistic and cultural guidelines for administration of the Preschool Language Scales, Fifth Edition (PLS5) in Arab-American preschool children, the normative sample curves should show the same distribution as a monolingual population, which would indicate that the test is valid. The test would be considered reliable if the results obtained were consistent across different sampling sites.

What tense is used to write the methods section? +

The methods section is written in the past tense because it describes what was done.

Recommended programs include Statistical Analysis Software (SAS) ,  Statistical Package for the Social Sciences (SPSS) ,  JMP ,  R software,  MATLAB , Microsoft Excel,  GraphPad Prism , and  Minitab .

One of the most important aspects of a thesis, dissertation or research paper is the correct formulation of the aims and objectives. This is because your aims and objectives will establish the scope, depth and direction that your research will ultimately take. An effective set of aims and objectives will give your research focus and your reader clarity, with your aims indicating what is to be achieved, and your objectives indicating how it will be achieved.

Introduction

There is no getting away from the importance of the aims and objectives in determining the success of your research project. Unfortunately, however, it is an aspect that many students struggle with, and ultimately end up doing poorly. Given their importance, if you suspect that there is even the smallest possibility that you belong to this group of students, we strongly recommend you read this page in full.

This page describes what research aims and objectives are, how they differ from each other, how to write them correctly, and the common mistakes students make and how to avoid them. An example of a good aim and objectives from a past thesis has also been deconstructed to help your understanding.

What Are Aims and Objectives?

Research aims.

A research aim describes the main goal or the overarching purpose of your research project.

In doing so, it acts as a focal point for your research and provides your readers with clarity as to what your study is all about. Because of this, research aims are almost always located within its own subsection under the introduction section of a research document, regardless of whether it’s a thesis , a dissertation, or a research paper .

A research aim is usually formulated as a broad statement of the main goal of the research and can range in length from a single sentence to a short paragraph. Although the exact format may vary according to preference, they should all describe why your research is needed (i.e. the context), what it sets out to accomplish (the actual aim) and, briefly, how it intends to accomplish it (overview of your objectives).

To give an example, we have extracted the following research aim from a real PhD thesis:

Example of a Research Aim

The role of diametrical cup deformation as a factor to unsatisfactory implant performance has not been widely reported. The aim of this thesis was to gain an understanding of the diametrical deformation behaviour of acetabular cups and shells following impaction into the reamed acetabulum. The influence of a range of factors on deformation was investigated to ascertain if cup and shell deformation may be high enough to potentially contribute to early failure and high wear rates in metal-on-metal implants.

Research Objectives

Where a research aim specifies what your study will answer, research objectives specify how your study will answer it.

They divide your research aim into several smaller parts, each of which represents a key section of your research project. As a result, almost all research objectives take the form of a numbered list, with each item usually receiving its own chapter in a dissertation or thesis.

Following the example of the research aim shared above, here are it’s real research objectives as an example:

Example of a Research Objective

  • Develop finite element models using explicit dynamics to mimic mallet blows during cup/shell insertion, initially using simplified experimentally validated foam models to represent the acetabulum.
  • Investigate the number, velocity and position of impacts needed to insert a cup.
  • Determine the relationship between the size of interference between the cup and cavity and deformation for different cup types.
  • Investigate the influence of non-uniform cup support and varying the orientation of the component in the cavity on deformation.
  • Examine the influence of errors during reaming of the acetabulum which introduce ovality to the cavity.
  • Determine the relationship between changes in the geometry of the component and deformation for different cup designs.
  • Develop three dimensional pelvis models with non-uniform bone material properties from a range of patients with varying bone quality.
  • Use the key parameters that influence deformation, as identified in the foam models to determine the range of deformations that may occur clinically using the anatomic models and if these deformations are clinically significant.

It’s worth noting that researchers sometimes use research questions instead of research objectives, or in other cases both. From a high-level perspective, research questions and research objectives make the same statements, but just in different formats.

Taking the first three research objectives as an example, they can be restructured into research questions as follows:

  • Can finite element models using simplified experimentally validated foam models to represent the acetabulum together with explicit dynamics be used to mimic mallet blows during cup/shell insertion?
  • What is the number, velocity and position of impacts needed to insert a cup?
  • What is the relationship between the size of interference between the cup and cavity and deformation for different cup types?

Difference Between Aims and Objectives

Hopefully the above explanations make clear the differences between aims and objectives, but to clarify:

  • The research aim focus on what the research project is intended to achieve; research objectives focus on how the aim will be achieved.
  • Research aims are relatively broad; research objectives are specific.
  • Research aims focus on a project’s long-term outcomes; research objectives focus on its immediate, short-term outcomes.
  • A research aim can be written in a single sentence or short paragraph; research objectives should be written as a numbered list.

How to Write Aims and Objectives

Before we discuss how to write a clear set of research aims and objectives, we should make it clear that there is no single way they must be written. Each researcher will approach their aims and objectives slightly differently, and often your supervisor will influence the formulation of yours on the basis of their own preferences.

Regardless, there are some basic principles that you should observe for good practice; these principles are described below.

Your aim should be made up of three parts that answer the below questions:

  • Why is this research required?
  • What is this research about?
  • How are you going to do it?

The easiest way to achieve this would be to address each question in its own sentence, although it does not matter whether you combine them or write multiple sentences for each, the key is to address each one.

The first question, why , provides context to your research project, the second question, what , describes the aim of your research, and the last question, how , acts as an introduction to your objectives which will immediately follow.

Explaining aims vs objectives

Note: Your research aims need not be limited to one. Some individuals per to define one broad ‘overarching aim’ of a project and then adopt two or three specific research aims for their thesis or dissertation. Remember, however, that in order for your assessors to consider your research project complete, you will need to prove you have fulfilled all of the aims you set out to achieve. Therefore, while having more than one research aim is not necessarily disadvantageous, consider whether a single overarching one will do.

Research Objectives

Each of your research objectives should be SMART :

  • Specific – is there any ambiguity in the action you are going to undertake, or is it focused and well-defined?

Mistakes in Writing Research Aims and Objectives

1. making your research aim too broad.

Having a research aim too broad becomes very difficult to achieve. Normally, this occurs when a student develops their research aim before they have a good understanding of what they want to research. Remember that at the end of your project and during your viva defence , you will have to prove that you have achieved your research aims; if they are too broad, this will be an almost impossible task. In the early stages of your research project, your priority should be to narrow your study to a specific area. A good way to do this is to take the time to study existing literature, question their current approaches, findings and limitations, and consider whether there are any recurring gaps that could be investigated .

Note: Achieving a set of aims does not necessarily mean proving or disproving a theory or hypothesis, even if your research aim was to, but having done enough work to provide a useful and original insight into the principles that underlie your research aim.

2. Making Your Research Objectives Too Ambitious

Be realistic about what you can achieve in the time you have available. It is natural to want to set ambitious research objectives that require sophisticated data collection and analysis, but only completing this with six months before the end of your PhD registration period is not a worthwhile trade-off.

3. Formulating Repetitive Research Objectives

Each research objective should have its own purpose and distinct measurable outcome. To this effect, a common mistake is to form research objectives which have large amounts of overlap. This makes it difficult to determine when an objective is truly complete, and also presents challenges in estimating the duration of objectives when creating your project timeline. It also makes it difficult to structure your thesis into unique chapters, making it more challenging for you to write and for your audience to read.

The introduction: The introduction is the first section and it provides as the name implies an introduction to the thesis. The introduction contains such aspects as the background to the study which provides information on the topic in the context of what is happening in the world as related to the topic. It also discusses the relevance of the topic to society, policies formulated success and failure. The introduction also contains the statement of the problem which is essentially a succinct description of the problem that the thesis want to solve and what the trend will be if the problem is not solved. The concluding part of the statement of problem ends with an outline of the research questions. These are the questions which when answered helps in achieving the aim of the thesis. The third section is the outline of research objectives. Conventionally research objectives re a conversion the research questions into an active statement form. The main body includes the following; the literature review, methodology, research results and discussion of the result, the summary, conclusion and recommendations, the list of references and the appendices.

The literature review : The literature review is often the most voluminous aspects of a thesis because it reviews past empirical and theoretical literature about the problem being studied. This section starts by discussing the concepts relevant to the problem as indicated in the topic, the relationship between the concepts and what discoveries have being made on topic based on the choice of methodologies. The validity of the studies reviewed are questioned and findings are compared in order to get a comprehensive picture of the problem. The literature review also discusses the theories and theoretical frameworks that are relevant to the problem, the gaps that are evident in literature and how the thesis being written helps in resolving some of the gaps.

The major importance of Literature review is that it specifies the gap in the existing knowledge (gap in literature). The source of the literature that is being reviewed should be specified. For instance; ‘It has been argued that if the rural youth are to be aware of their community development role they need to be educated’ Effiong, (1992). The methodology: The third section is a discussion of the research methodology adopted in the thesis and touches on aspects such as the research design, the area, population and sample that will be considered for the study as well as the sampling procedure. It is the chapter 3. This chapter explains the method used in data collection and data analysis. The findings and discussion of result : The next section is a discussion of findings based on the data collection instrumentation used and the objectives or hypotheses of study if any. It is the chapter 4. It is research results. This is the part that describes the research. It shows the result gotten from data that is collected and analysed. Summary, Conclusion and Recommendation: This is normally the chapter 5. The last section discusses the summary of the study and the conclusions arrived at based on the findings discussed in the previous section. References: It cite all ideas, concepts, text, data that are not your own. It is acceptable to put the initials of the individual authors behind their last names. The way single author is referenced is different from the way more than one author is referenced (RE: write a thesis or writing a thesis).

The appendices; it includes all data in the appendix. Reference data or materials that is not easily available. It includes tables and calculations, List of equipment used for an experiment or details of complicated procedures. If a large number of references are consulted but all are not cited, it may also be included in the appendix. The appendices also contain supportive or complementary information like the questionnaire, the interview schedule, tables and charts while the references section contain an ordered list of all literature, academic and contemporary cited in the thesis. Follow the following steps to achieve successful thesis writing

Start writing early. Do not delay writing until you have finished your project or research. Write complete and concise “Technical Reports” as and when you finish each nugget of work. This way, you will remember everything you did and document it accurately, when the work is still fresh in your mind. This is especially so if your work involves programming.

Spot errors early. A well-written “Technical Report” will force you to think about what you have done, before you move on to something else. If anything is amiss, you will detect it at once and can easily correct it, rather than have to re-visit the work later, when you may be pressured for time and have lost touch with it.

Write your thesis from the inside out. Begin with the chapters on your own experimental work. You will develop confidence in writing them because you know your own work better than anyone else. Once you have overcome the initial inertia, move on to the other chapters.

End with a bang, not a whimper. First things first, and save the best for last. First and last impressions persist. Arrange your chapters so that your first and last experimental chapters are sound and solid.

Write the Introduction after writing the Conclusions. The examiner will read the Introduction first, and then the Conclusions, to see if the promises made in the former are indeed fulfilled in the latter. Ensure that your introduction and Conclusions match.

“No man is an Island”. The critical review of the literature places your work in context. Usually, one third of the PhD thesis is about others’ work; two thirds, what you have done yourself. After a thorough and critical literature review, the PhD candidate must be able to identify the major researchers in the field and make a sound proposal for doctoral research. Estimate the time to write your thesis and then multiply it by three to get the correct estimate. Writing at one stretch is very demanding and it is all too easy to underestimate the time required for it; inflating your first estimate by a factor of three is more realistic.

Punctuating your thesis

Punctuation Good punctuation makes reading easy. The simplest way to find out where to punctuate is to read aloud what you have written. Each time you pause, you should add a punctuation symbol. There are four major pause symbols, arranged below in ascending order of “degree of pause”:

  • Comma. Use the comma to indicate a short pause or to separate items in a list. A pair of commas may delimit the beginning and end of a subordinate clause or phrase. Sometimes, this is also done with a pair of “em dashes” which are printed like this:
  • Semi-colon. The semi-colon signifies a longer pause than the comma. It separates segments of a sentence that are “further apart” in position, or meaning, but which are nevertheless related. If the ideas were “closer together”, a comma would have been used. It is also used to separate two clauses that may stand on their own but which are too closely related for a colon or full stop to intervene between them.
  • Colon. The colon is used before one or more examples of a concept, and whenever items are to be listed in a visually separate fashion. The sentence that introduced the itemized list you are now reading ended in a colon. It may also be used to separate two fairly—but not totally—independent clauses in a sentence.
  • Full stop or period. The full stop ends a sentence. If the sentence embodies a question or an exclamation, then, of course, it is ended with a question mark or exclamation mark, respectively. The full stop is also used to terminate abbreviations like etc., (for et cetera), e.g., (for exempli gratia), et al., (for et alia) etc., but not with abbreviations for SI units. The readability of your writing will improve greatly if you take the trouble to learn the basic rules of punctuation given above.

Humanities theses are often structured more like a longer-form essay . Just like in an essay, you build an argument to support a central thesis.

An introduction sets up the topic, purpose, and relevance of your thesis, as well as expectations for your reader. This should:

  • Ground your research topic , sharing any background information your reader may need
  • Define the scope of your work
  • Introduce any existing research on your topic, situating your work within a broader problem or debate
  • State your research question(s)
  • Outline (briefly) how the remainder of your work will proceed

Theoretical framework

Your literature review can often form the basis for your theoretical framework, but these are not the same thing. A theoretical framework defines and analyses the concepts and theories that your research hinges on.

Your methodology chapter shows your reader how you conducted your research. It should be written clearly and methodically, easily allowing your reader to critically assess the credibility of your argument. Furthermore, your methods section should convince your reader that your method was the best way to answer your research question.

A methodology section should generally include:

  • Your overall approach ( quantitative vs. qualitative )
  • Your research methods (e.g., a longitudinal study )
  • Your data collection methods (e.g., interviews or a controlled experiment
  • Any tools or materials you used (e.g., computer software)
  • The data analysis methods you chose (e.g., statistical analysis , discourse analysis )
  • A strong, but not defensive justification of your methods

Your results section should highlight what your methodology discovered. These two sections work in tandem, but shouldn’t repeat each other. While your results section can include hypotheses or themes, don’t include any speculation or new arguments here.

Your discussion section is where you can interpret your results in detail. Did they meet your expectations? How well do they fit within the framework that you built? You can refer back to any relevant source material to situate your results within your field, but leave most of that analysis in your literature review.

Your thesis conclusion should concisely answer your main research question. It should leave your reader with an ultra-clear understanding of your central argument, and emphasise what your research specifically has contributed to your field.

Once you’re done writing, the next part of your editing process begins. Leave plenty of time for proofreading and editing prior to submission. Nothing looks worse than grammar mistakes or sloppy spelling errors!

Once you’ve submitted your final product, it’s common practice to have a thesis defense, an oral component of your finished work. This is scheduled by your advisor or committee, and usually entails a presentation and Q&A session.

After your defense, your committee will meet to determine if you deserve any departmental honors or accolades. However, keep in mind that defenses are usually just a formality. If there are any serious issues with your work, these should be resolved with your advisor way before a defense.

The conclusion of your thesis or dissertation shouldn’t take up more than 5-7% of your overall word count.

When you mention different chapters within your text, it’s considered best to use Roman numerals for most citation styles. However, the most important thing here is to remain consistent whenever using numbers in your dissertation .

If you only used a few abbreviations in your thesis or dissertation, you don’t necessarily need to include a list of abbreviations .

If your abbreviations are numerous, or if you think they won’t be known to your audience, it’s never a bad idea to add one. They can also improve readability, minimising confusion about abbreviations unfamiliar to your reader.

A thesis or dissertation outline is one of the most critical first steps in your writing process. It helps you to lay out and organise your ideas and can provide you with a roadmap for deciding what kind of research you’d like to undertake.

Generally, an outline contains information on the different sections included in your thesis or dissertation, such as:

  • Your anticipated title
  • Your abstract
  • Your chapters (sometimes subdivided into further topics like literature review, research methods, avenues for future research, etc.)

What is a thesis statement.

A thesis statement is a sentence that states the main idea of your paper. It is not just a statement of fact, but a statement of position. What argument are you making about your topic? Your thesis should answer that question.

How long should my thesis statement be?

Thesis statements are often just one sentence. Keep thesis statements concise, without extra words or information. If you are having trouble keeping your thesis statement to one sentence, consider the following:

  • Is your thesis is specific enough?
  • Does your thesis directly supports your paper?
  • Does your thesis accurately describes your purpose or argue your claim?

Honors Program - Senior Thesis

The Honors Program is open to all majors with a GPA of 3.5 in History, and centers around an original piece of research and writing undertaken with the guidance of a thesis advisor. The six-credit program involves a seminar, one-on-one consultations with the advisor, and a 50-60 page thesis.

Eligible seniors have the opportunity to write a 12-credit thesis (counting as four upper-division electives) as a Scholar of the College. The Scholar of the College program follows the same timeline, but the thesis requirements are more extensive and the thesis involves a more ambitious project.

If you are hoping to go to graduate school in the humanities, you should seriously consider writing a thesis because it will give you a sense of the kind of work that will be expected at that level.

But you need not have graduate school in mind to write a thesis. If you want to bring all of your undergraduate training to bear on a single, challenging project the honors program is for you. The thesis experience, picking a topic, researching it and producing an original piece of writing will challenge you in ways you have not yet been challenged. It is also incredibly rewarding. Working with other seniors and one-on-one with award winning faculty is a once in a lifetime experience. Such an accomplishment is the culmination of a liberal arts education and will set you up for a lifetime of achievement.

What does the thesis look like at the end of the process?

The thesis itself should be about 50-60 pages long and generally includes multiple chapters. It should be bound in a clear plastic binder that will both hold the pages securely and permit the title page to be read without opening it. Plastic binders are available at most stationary stores. The thesis alone is an impressive accomplishment but you can also take advantage of further publishing opportunities. There are two different kinds of publishing opportunity. The first and easiest is to deposit it with the Boston College eScholarship repository. The second involves an application and editing process to an undergraduate journal. This process is more involved than asimply placing your thesis in a repository, but it is more prestigious and has the potential to reach more readers.

Columbia Undergraduate Journal of History

The Columbia Undergraduate Journal of History the United States' leading undergraduate social science research journal. We publish papers from all social science disciplines, including political science, economics, sociology, history, psychology, linguistics, law, anthropology, criminology, cultural and area studies, development studies, and demography. 

Elements was founded in September 2004 by a group of twenty undergraduate students. The journal published its first issue in May of 2005, featuring research articles written by Boston College undergraduates along with shorter special features. The goal of the publication is to become a forum for the exchange of original ideas within and across disciplines at the university. Staff members read and evaluate all submitted manuscripts and select the best articles on the basis of quality of scholarship as well as readability. Ezra's Archives is a publication put forth annually by the Cornell Historical Society. This journal, launched in the Spring of 2011, showcases stellar examples of undergraduate research in the field of history. History Matters, Appalachian State University

This undergraduate History Matters history journal is published annually on a website by the History Department at Appalachian State University. The journal is edited by undergraduates with the help of a faculty board. All submissions and editing must be completed while authors are still undergraduates.

Journal of Undergraduate International Studies, University of Wisconsin The Journal of Undergraduate International Studies at the University of Wisconsin-Madison invites unpublished reflections on a wide range of academic and creative disciplines from contributors worldwide. Submitting authors should be within one semester of graduation. Topics include, but are not limited to, international conflict and resolution environmental issues, economics, development and trade, global security and international health.

The Yale Historical Review

The Yale Historical Review provides undergraduates an opportunity to have their exceptional work highlighted and encourages the diffusion of original historical ideas on campus by providing a forum for outstanding undergraduate history papers covering any historical topic. Students may submit history papers they've written over the course of their college careers.

Report, United States Military Academy

Report  is a historical review that publishes the work of undergraduate students. They are expanding to include submissions from students at other colleges and universities. They encourage submissions from all undergraduates interested in historical research. Report is published both electronically and in booklet form during the spring and fall semesters. Thesis research projects, extracellular vesicles in health and disease.

Mentors:   Tom Gallagher, PhD and Gopal Gupta, MD

Research Interest :    Our primary research interests are in virology.  We specifically focus our attention on entry and pathogenesis of enveloped viruses, particularly human pathogenic coronaviruses, such as the Severe Acute Respiratory Syndrome (SARS) and Middle East Respiratory Syndrome (MERS) coronaviruses. We also study transmission of extracellular vesicles and compare and contrast how enveloped virus and extracellular vesicle mediated the transfer of cytoplasmic contents between cells.

Extracellular Vesticles in Health and Disease

Extracellular vesicles (EVs) are ubiquitous, submicroscopic, membrane-delimited particles that are continuously secreted from both healthy and diseased cells.  They accumulate in bodily fluids and may be useful “biomarkers” of patient health status.  While the detailed structures and functions of EVs remain obscure, recent findings indicate that EVs transmit RNAs and proteins between cells in the body, and therefore are agents of intercellular communication.  These intercellular transfers may be clinically relevant – indeed there is compelling evidence that EVs have therapeutic potential in cardiovascular disease.  We have developed sensitive assays to measure EV-directed intercellular protein transfers.  Projects include:  (1)  identifying EV components facilitating or restricting intercellular transfers,  (2)  characterizing the tissues and cells targeted by transfer-competent EVs, and  (3) determining the relationships between EV cargo delivery into target cells and EV-mediated therapeutic activities in mouse models of human disease.  These projects link EV molecular cellular biology investigations with those aimed at evaluating EVs as potential therapeutic agents.

Decontamination of Clostridium Difficile Spores

Collaborators: Adam Driks, Ph.D. and Stuart Johnson, M.D.

Clostridium difficile  contamination is widespread in hospitals, significantly increasing the risk of infection for susceptible patients. We seek to reduce this risk by developing chemistries that remove  C. difficile  from hospital environments which are safe and easy to apply, and do not present additional risks to patients.

The Microbiome of Ocular Surface Diseases

We are studying the ocular microbiome and its role in floppy eyelid disease (FES), Steven’s Johnson Syndrome (SJS), graph vs. host disease (GVHD) and dry eye disease (DED).  The eye contains few bacteria compared to other parts of the body, but we have developed robust methods to interrogate the microbiome in this body site. Preliminary results have found that normal eyes tend to have a  Lactobacillus/Streptococcus  microbiome or a microbiome with lots of  Corynebacterium.  We have found that patients with SJS have much more  Staphylococcus  than other patients or healthy eyes, while GVHD patients look similar to healthy individuals and FES and DED are dominated by  Corynebacterium.  We are currently studying the viability and stability of these microbiomes longitudinally.

Urinary Microbiome

We have projects working on the urinary microbiome in healthy women and women with bladder, vaginal or kidney disorders.  Most women have  Lactobacillus  in their urine and most women’s bladders have a dominant bacterial genera that accounts for >50% of the bacteria that are detected.  We have also found that many bacterial genera can be present, such as  Staphylococcus , but at a much lower frequency.  In chronic kidney disease (CKD), we have found that CKD patients have more diverse microbiomes than other patients. This may explain why these patients are more susceptible to infections and other clinical outcomes, and we are currently exploring those opportunities.

Association of NPC1L1L Polymorphisms with HCV Infection Outcome

Mentors:  Susan Uprichard, PhD and Scott Cotler, MD

Genetic polymorphisms within IL28B are associated with HCV clearance and treatment response; however, these associations do not account for all the variability in infection and treatment response. We identified the host cell cholesterol uptake receptor NPC1L1 as an HCV entry factor. HCV sensitivity to NPC1L1 inhibition correlates with cholesterol levels within the viral particle. If HCV uptake is mediated by NPC1L1 cholesterol uptake activity, then NPC1L1 SNPs associated with high/low cholesterol absorption may also affect HCV infections. Thus, we are studying the impact of NPC1L1 genetic polymorphisms on cell permissiveness to HCV infection as well as infection and treatment outcomes. We want to determine if NPC1L1 SNPs alter permissiveness of cells to HCV infection and /or  correlate with HCV spontaneous clearance, and/or treatment response.

Clostridium Difficile Ecology in the Gut

Collaborators: Adam Driks, PhD and Dale Gerding, MD

Ingestion of C. difficile spores is critical to establishing an infection, yet the fate of the spore in the gut and its role in adhesion to the gut epithelium is unknown. It is very likely that C. difficile cells exist within a biofilm in the gut. To begin to address this question, we will search for conditions in vitro that stimulate C. difficile biofilm formation, and characterize any resulting extracellular matricies.

Colonization of the gut by C. difficile spores is critical to establishment of disease. Possibly, it is also important in disease recurrence, a growing problem in hospitals. Persistence of C. difficile spores in the gut could explain recurrence. To begin to address this possibility, we will analyze the molecular basis of spore-epithelium interactions.

Mechanisms Streptococcus Pneumoniae Pathogensis

Mentors:  Andrew T. Ulijasz, PhD and Clinician TBD

Gram-positive pathogen  Streptococcus pneumoniae  is responsible for 25% of all preventable deaths in children under the age of 5, and the leading cause in the US of community acquired pneumonia-related deaths. Although  S. pneumoniae  is responsible for a considerable disease burden on society, it first resides as a harmless commensal in the nasopharynx, before disseminating into the host to cause a variety of infections under situations where one’s immune system is compromised. A key component of the success of the pneumococcus is its ability to avoid innate immunity via its polysaccharide capsule, which is its major virulence factor and defines the > 90 capsule-specific serotypes. As the current vaccine is composed only of a subset of these serotypes, it would be beneficial to identify new therapeutics for targeting all pneumococcal serotypes rather than a few. The goal of our research is to understand conserved molecular events that trigger the deadly switch from commensal to pathogenesis. Through this research we have discovered a novel pathway that we think is responsible for this key change in carrier state and involving capsule tissue-specific regulation. Excitingly, it appears this system is a novel macromolecular stressosome structure conserved in Gram-positive pathogens.

Project 1: Use ChIPseq next generation sequencing and standard biochemical methods to determine the regulation of SpxR, the key transcription factor in the pathway.

Project 2: Use super resolution microscopy and FRET to determine the “when and where” the signaling system is utilized.

Project 3: Determine the substrate binding specificity of the signaling complex using genetic and biochemical techniques.

Pathogenesis of HPV: Defining Viral and Host Factors that Determine Progression

Mentors:  Susan Uprichard, PhD, James Cook, MD, Gail Reaid, MD, and Nina Clark, MD

Cancer is associated with human papillomavirus infection (HPV), the most common sexually transmitted infection in the US. There has been an increase in the incidence of HPV cancer in recent years but there are multiple questions that remain to be elucidated about the pathogenesis of HPV-associated cancer including the natural history of precursor anal dysplastic lesions, the risks associated with the development and progression of dysplasia, the contribution of various HPV serotypes to the development of cancer and the immunologic events that may impact progression of anal dysplasia to cancer.  We are therefore studying persons at high risk for HPV-related cancer to assess the prevalence of anal neoplasia, and study host and viral factors associated with its development. Specifically, we obtain pap smears from high-risk persons to determine the prevalence of abnormal cytology, assess the subtypes of HPV present, and determine potential associations of demographic and clinical factors with the presence of dysplasia.  Long term, we will then follow the patients with dysplasia to assess changes over time and assess whether microRNA profiles in blood or dysplastic tissue are associated with progression to cancer.  These studies should provide a better understanding of the factors that contribute to the development of cancer and enable early recognition and management.

Determining the Mechanism by which Probiotics Inhibit Inflammation

Mentors: Katherine L. Knight, PhD and TBD clinician

Macrophages are critical immune mediators and key players in the clearance of viral infections.  This project will investigate how the model coronavirus, mouse hepatitis virus, affects the phagocytic activity of macrophages.  Students will learn how to: isolate and propagate bone marrow-derived macrophages; analyze transcriptional activation in mock-infected and virus-infected macrophages; and evaluate how viral factors modulate the innate immune response in activated macrophages.

Human Bladder Epithelial Culture System for Studying Host Pathogen...

Mentors: Phong T. Le, PhD, Beth Mueller, MD, FACS, MS

The human urinary tract (UT) provides critical mechanical barriers to infection; however, UT is one of the most common sites for bacterial infection. UT infections (UTIs) are initiated when uropathogenic Escherichia. coli. (UPEC) invade and colonize the superficial epithelial or bladder epithelial cells – the urothelial bladder cells (BUC) that line the bladder lumen. Entrance into BUCs allows UPEC to proliferate and establish intracellular bacterial communities. In response to the invasion and colonization, BUC responds by mounting innate and adaptive immune response leading to exfoliation of infected cells. This project will utilize primary human BUC to interrogate its interaction with host immune cells of the bladder, BUC-UPEC interactions and mechanisms by which UPEC escape immune response and cause recurrences of UTI.  

Characterization of Human Bidirectional T-Cells

Mentors: Makio Iwashima, PhD, Paula White, MD and Paul Jones MD, FACS

Infants have a suppressed immune system which makes them highly vulnerable to infections and limits their immune responses to protective and life-saving vaccines.  Both adaptive and innate immune responses in infant are less effective than those in adult.  To provide better health care for infants, understanding the cellular and molecular basis of how the ho infant immune system works is essential.

In our recent studies, we determine that antigen receptor stimulation of umbilical cord blood T cells promotes development of Foxp3+ T cells that are immunosuppressive and also express pro-inflammatory cytokines.  Gene expression profiles show these T cells are distinctive group of T cells from previously known regulatory or effector T cells.  Thus, we termed these T cells “Bi directional T cells” (BiT cells).  In this study, we will determine if this unique group of Foxp3+ T cells are present in the children and adult human tissues and peripheral blood under normal conditions and test their functions.

Microbiome of the Urinary Tract

The current paradigm for urinary tract infections and other lower urinary tract disorders does not take into account evolving knowledge of the human urobiome, the microbial communities of the lower urinary tract. The NIH-funded Human Microbiome Project has shown that microbiome composition contributes to diverse health and disease states. Yet, these studies did not report on the urobiome, largely because reviewers believed the “bladder is sterile” dogma, which results primarily from use of standard urine culture-dependent methods that were optimized to detect E. coli. High throughput 16S rRNA gene sequencing and an enhanced urine culture method that we call Expanded Quantitative Urine Culture (EQUC) reveal bacterial DNA and live bacteria, respectively, in standard urine culture-negative samples. These studies have revealed associations between the female urobiome and post-instrumentation and post-operative UTI, urgency urinary incontinence (UUI) and response to overactive bladder treatment. Some  Lactobacillius  species are associated with lack of symptoms and protection against post-instrumentation UTI. These results are consistent with the probability that the human urobiome has protective capabilities and that disruption (dysbiosis) can result in disorders such as UTI.

The Loyola Urinary Education and Research Collaborative (LUEREC), co-directed by Drs. Wolfe and Mueller, is a trans-disciplinary translational research team composed of clinicians, basic scientists, clinical microbiologists, and members of both the Loyola Genomics Facility & the Center for Bioinformatics. LUEREC wants to know how the urobiome affects the health of women and men. Our primary hypothesis is that bacterial biodiversity exists in the bladders of normal women & that perturbations in this normal bacterial biodiversity may contribute to symptoms. In simpler terms, we wish to learn the answers to the following questions: Which bacteria are present in the bladder? How does the composition of the bladder microbiome compare to the microbiomes of the surrounding organs or surfaces, e.g. the urethra, vagina or perineum? Which bacteria are protective? Which ones are pathogenic?

Ongoing studies are designed to:  (1)  determine if EQUC outperforms the standard urine culture as a diagnostic tool for urinary tract infections,  (2)  distinguish the microbiome of the female bladder from that of the female urethra,  (3)  understand the daily dynamics of the urobiome in young healthy women,  (4)  determine when the urobiome becomes established in children,  (5)  characterize the urobiome of women with recurrent urinary tract infections and follow the effects of antibiotic treatments,  (6)  investigate the role of the urobiome in the development of kidney stones,  (7)  learn how members of the urobiome interact with each other, and (8) understand understudied uropathogens. We anticipate that the totality of these and other studies will facilitate a paradigm change in our understanding of urinary tract in health and disease.

Complement Component C4 in the Pathogenesis OF Sjögren’s syndrome

Mentors:  Yee Ling Wu, PhD, Rochella Ostrowski, MD and Rodney Tehrani, MD

Sjögren’s syndrome is a systemic autoimmune disease with overlapping clinical features with diseases such as lupus. Low levels of complement C4 protein are often observed in Sjögren’s syndrome, and are associated with cryoglobulinemia and lymphoma. Genetic deficiency and low gene copy numbers of complement C4 have been shown to be an important risk factor for lupus. Activation of C4 protein during inflammation consumes C4 protein and mediates tissue injuries. We hypothesize that C4 genetic and protein polymorphisms are engaged in disease predisposition and in modulating the clinical presentation of Sjögren’s syndrome. We will analyze patient samples using accurate molecular assays for determining C4 gene copy numbers, immunoassays for activated complement protein products, and transcriptomic analyses to i) determine the cause of low C4 in Sjögren’s syndrome, and to ii) evaluate the utility of combining genetic stratification of C4 and new protein markers in the diagnosis and management of Sjögren’s syndrome.

EPIC in the US: Is It Pathogenic?

Mentors: Gail Hecht and Neil Gupta, MD

A new comprehensive and rapid multiplex PCR assay for 22 enteric pathogens has revealed that enteropathogenic  E. coli  (EPEC), previously unrecognized as a common pathogen in the USA, is frequently identified in patients with diarrhea in the USA and even in healthy adults without diarrhea. This unexpected finding presents confusion regarding the role of EPEC in disease, therefore warranting investigation into the virulence properties of these strains. This project will isolate and characterize these clinical EPEC strains to provide insight into their pathogenic potential and thus help to inform clinical care of patients harboring these organisms.

The Viral Response to Antiviral Therapies

Mentors:  Bryan Mounce, PhD and Gail Reid, MD

Viruses notoriously gain resistance to antivirals, owing to their high mutation rates and rapid replication strategies. We are interested in understanding the molecular determinants of how viruses overcome antivirals, considering what changes occur to the viral genome, thus affecting the viral proteins, replication, and pathogenesis. We can use this information to understand how viruses infect a cell and replicate, as well as how we can prevent the emergence of antiviral resistance.

Genetic Manipulation of Potential Uropathogens

Mentors:  Karen Visick, PhD and Amanda Harrington, MD

Until recently, urine was believed to be sterile. Now, work from Loyola, in particular from the Wolfe lab, has demonstrated the existence of numerous microbes in urine. Some of these microbes may be “normal flora” bacteria, while others have been associated with the disease state. In collaboration with the Wolfe lab, our long term goals seek to understand the roles of specific urinary isolates in disease. To achieve these goals, we propose first to develop tools for the genetic manipulation of the urinary isolates. Then, we will mutate specific genes with the potential to be involved in virulence. Finally, through collaborations, we will develop assays of virulence to determine if the mutant bacteria exhibit reduced or loss of virulence.

Determining how Variations in Virulence Factor Regulation Enhance the Pathogenesis of Clinical Isolates of S.Aureus

Research Interests :  We investigate the molecular mechanisms underlying  Staphylococcus aureus  disease processes and bacterial evasion of the host immune response.  For a bacterial pathogen to cause disease it often must: acquire metabolic resources from nutrient restricted environments; escape host immune defenses; produce virulence factors that damage host cells or usurp host functions; and/or use physical means to persist in inhospitable conditions. My laboratory aims to develop a better understanding of these pathogenic mechanisms in the bacterium  Staphylococcus aureus .

Polyamines in Cellular Processes

Mentors:  Bryan Mounce, PhD and Nina Clark, MD

During viral infection, the interferon response is triggered to limit viral replication. One way the cell limits viral replication is by depleting polyamines, through the enzyme SAT1. SAT1 acetylates polyamines, reversing their charges and clearing them from the cell. The effect this has on the cell isn’t known, however. Thus, we are interested in determining how polyamines may impact cellular signaling and what impact this has on replication of diverse viruses.

Polyamines in Viral Processes

Viruses rely on polyamines for translation of the viral genomes and to stimulate the RNA polymerase. However, precisely how polyamines are involved in these processes isn’t entirely understood. We are working to understand how polyamines affect these processes in diverse viruses like Zika virus, Coxsackievirus, enterovirus, and Rift Valley fever virus. Understanding how viruses rely on polyamines can inform how we can better treat viral infection with inhibitors, develop new inhibitors, or use combination therapies. We are also interested in how viruses evolve to overcome polyamine depletion, which has important implications for antiviral resistance.

Disecting HIV-Host Interactions

Mentors:  Edward Campbell, PhD, and Clinician TBD

We are interested in understanding the interactions occurring between HIV-1 and target cells that lead to, or prevent, productive infection. This includes understanding how cellular proteins, termed restriction factors, can inhibit HIV-1 infection, and how HIV-1 and other retroviruses can evade these restriction factors to replicate efficiently. We are also working to understand how HIV-1 interacts with cellular proteins to facilitate infection, specifically examining how HIV-1 engages the microtubule cytoskeleton and nuclear pore complex during infection.

Understanding the Mechanism of Amyloid Protein Transmission

Mentors:  Edward Campbell, PhD and Clinician TBD

We are interested in understanding the “virus like” transmission of amyloid proteins that occur in neurodegenerative diseases. By understanding the mechanism by which these proteins, such as alpha-synuclein in Parkinson’s disease, spread from cell to cell, we hope to identify opportunities to arrest disease development in patients with Parkinson’s disease, Alzheimer’s disease and Huntington’s disease. We are currently examining how these amyloid proteins enter neurons and induce cellular dysfunction that ultimately drives the continued spread of these amyloid proteins and their associated pathology.

Inflammation Biosensor Development

We are interested in understanding how inflammation drives the pathogenesis of numerous human diseases. To this end, we have developed a novel biosensor which can monitor inflammation in mouse models of human disease. The ability to monitor inflammation in living animals may provide experimental tools with which to better understand and treat numerous human diseases.

Macaulay Honors College

The 2024 macaulay honors college outstanding thesis and capstone awards, college news.

The inaugural Macaulay Honors College Outstanding Thesis and Capstone Awards recognize and reward excellence in undergraduate research, scholarship, or the arts. The consortium-wide competition aims to recognize excellence in undergraduate research, scholarship, or the arts by Macaulay Honors College seniors as evidenced through their theses and capstone projects. These awards also recognize the invaluable role of faculty mentors and advisors to thesis and capstone projects and to Macaulay students’ overall honors education.

Awards are given to the top three projects submitted each year. First-place winners receive $1,000; second place is awarded $500; and third place is awarded $250. This year’s winners will be featured on the Macaulay Honors College website, and acknowledged at the 2024 Macaulay Honors College Commencement Ceremony.

Student theses and capstone projects will be evaluated by a committee of Macaulay faculty and staff on the basis of the work’s quality, originality, and contribution to the field.

Eligibility

All Macaulay Honors College seniors completing a thesis or capstone project as part of their graduation requirements in the 2023-2024 academic year are eligible. The Spring 2024 award encompasses graduates from Summer and Fall 2023 as well as Spring 2024.

Students are invited to apply directly, and nominations from academic departments, individual faculty members, and campus Macaulay Directors are also encouraged. Students nominated by faculty and staff will need to complete the online application.

How to Apply

We are now accepting applications and nominations for the 2024 Macaulay Honors College’s Outstanding Thesis and Capstone Awards. Applications must be submitted by Thursday, May 2, 2024, 11:59pm EST. We encourage faculty, staff, and directors intending to nominate students to do so as soon as possible. In submitting their online entry form, students should:

  • Project description
  • Description of methodology
  • Key findings (if appropriate)
  • Conclusions and implications
  • List any awards or grants received to support this project
  • Consult their thesis or project advisor that they would like to apply for this award. Faculty advisors will receive a short feedback form to complete. The advisor must complete the statement of support for the committee to evaluate the project.
  • Attach an excerpt of 5-10 pages (1250-2500 words) of your thesis or project in .pdf format.

The buzz is back with the Graduate School’s annual Three-Minute Thesis (3MT) Competition this spring! Earlier this month, 42 graduate students rocked the stage in front of a live audience all vying for a chance to advance to the final round and win cash prizes.

A panel of esteemed University faculty and postdocs had the challenging task of judging this year’s preliminary event, evaluating students’ presentation skills and research content. If you are unfamiliar with 3MT, it is an annual spectacle where master’s and doctoral students are tasked with condensing their research into a lightning-fast, three-minute presentation with only a single slide. It is an adrenaline-fueled sprint through the world of academia!

Since 2015, the Graduate School has hosted this event, showcasing the power, beauty and brilliance of graduate education at the University. In addition, recent winners of this competition have gone on to compete, and place, in regional 3MT competitions putting the University on the map as a hotbed of intellectual prowess.

We are thrilled to announce this year’s 16 finalists (see below) and cannot wait for the final showdown. The 3MT final round of competition is set to take place on Thursday, April 11, at 7 p.m. in the Wells Fargo Auditorium at the Mathewson-IGT Knowledge Center. Students, family, faculty and community members are invited to join us and witness firsthand the awe-inspiring brilliance of our scholars. For those who cannot attend in person, the event will be live-streamed via Zoom so please register here on Formstack to receive the information.
